O Brother where art thou? vandt en Grammy Award for Årets Album i 2002, Grammy Award for Bedste Country Collaboration with Vocals (for sangeren Dan Tyminski, hvis stemme overdubbede George Clooney 's i filmen på "I Am a Man of Constant Sorrow", Nashville sangskriver Harley Allen, og Nashville Bluegrass Bands Pat Enright), og Grammy-prisen for bedste mandlige country-vokalpræstation for "O, Death" af Ralph Stanley.
Albummet vandt prisen for årets album (kun det andet soundtrack nogensinde) og prisen for årets single for "I Am a Man of Constant Sorrow" ved Country Music Association Awards. Det vandt også prisen for årets album ved den 37. Academy of Country Music Awards og tog 2 internationale Bluegrass Music Awards med hjem: Årets album og årets gospeloptagelse (for Alison Krauss og Gillian Welch på "I fly away").
